I would suggest making a PvP assassin and try it out. There decently more of a change I think than the other classes with is why I like them. It really hard to give you an answer on that I personally would rater play an assassin builds than a mesmer.

Im going to assume youre talking about pve here.
Really...I would say neither. Reason is because both mesmer and assassin are shunned in pve and hardly accepted into groups. Assassin is affected by this because its one of the new classes and most everyone that bought factions made a ritualist primary and an assassin primary.
Also, assassin primary is more of a pvp character in my opinion, as is mesmer. Better used against human opponents than mobs of AI.
Up to you though, but thats what Ive found while playing the game over the last week and the last year and a half.
The characters that I made for factions are Ranger/Assassin and Monk/Ritualist. Lets you get the skills while not being in the mainstream. Hell you could even make a Monk/Assassin and just use monk skills.
